Selective dendritic fluorescent sensors for Zn(II).

چکیده
A series of dendritic 8-hydroxyquinoline (8-HQ) and 5-dialkyl(aryl)aminomethyl-8-HQ derivatives were synthesized and their fluoroionophoric properties toward representative alkali, alkaline earth, group IIIA and transition metal ions were investigated. Among the selected ions, Zn(II) enhanced the fluorescence of N-di-(methoxycarbonylethyl)aminoethyl-3-[4-(8-hydroxyquinolin-5-ylmethyl)piperazin-1-yl]-propanoic amide] (7) by 31-fold, while Al(III) caused enhancement to some extent. The absence of any significant fluorescence enhancement by the other ions examined renders 7a highly useful Zn(II)-selective fluorescent sensor.

His-tags as Zn(II) binding motifs in a protein-based fluorescent sensor.
Fluorescent indicators that allow real-time imaging of Zn(II) in living cells are invaluable tools for understanding Zn(II) homeostasis. Genetically encoded sensors based on fluorescence resonance energy transfer between fluorescent protein domains have important advantages over synthetic probes. We discovered that hexahistidine tags have a strong tendency to dimerize upon binding of Zn(II) in ...

Chapter 1. An Introduction to Fluorescent Zn(II) Sensors and Their Applications in Biological Systems This chapter opens with an overview of the numerous roles of zinc in biology, with an emphasis on labile Zn(II), that is, loosely bound or kinetically available stores of the ion. The synthesis, characterization, and application of fluorescent sensors for Zn(II) detection constitute an active f...
متن کاملSubtle modification of 2,2-dipicolylamine lowers the affinity and improves the turn-on of Zn(II)-selective fluorescent sensors.
The spectroscopic and proton- and Zn(II)-binding properties of two new members of the Zinpyr family of fluorescent sensors are reported. In ZP1B and ZP3B, a (2-picolyl)(4-picolyl)amine (2,4-DPA) moiety is installed in place of the di(2-picolyl)amine (2,2-DPA) ligand used in the parent sensors ZP1 and ZP3. This modification has the benefit of both lowering the proton-induced turn-on at physiolog...
متن کاملAre two better than one? Comparing intermolecular and intramolecular indicator displacement assays in pyrophosphate sensors.
Peptide receptors with Zn(ii)-DPA units and a covalently bound fluorescent coumarin indicator on an oxazole-containing scaffold are shown to function as more selective pyrophosphate sensors than the analogous chemosensing ensembles in indicator displacement assays.
متن کاملBiochemistry of mobile zinc and nitric oxide revealed by fluorescent sensors.
Biological mobile zinc and nitric oxide (NO) are two prominent examples of inorganic compounds involved in numerous signaling pathways in living systems. In the past decade, a synergy of regulation, signaling, and translocation of these two species has emerged in several areas of human physiology, providing additional incentive for developing adequate detection systems for Zn(II) ions and NO in...
